The Duel on Zephyr One was an attempt by Melinda May to escape the Zephyr One to assist S.H.I.E.L.D., but was confronted by Enoch.

During the Battle at the Temple of the Forgotten, S.H.I.E.L.D. agent Melinda May was mortally wounded when Izel drove a sword through her stomach. Nevertheless, May survived the wound long enough to be placed into a Healing Pod which would cure her.[2] May remained unconscious in the pod for a while, but eventually awakened in the Zephyr One after the plane had been transported into year 1931.
Only Enoch was present to witness May's return to life. Fearing the consequences that the wound and the healing could have had on May, Enoch was instructed by Jemma Simmons to sedate her and bring her back into the Healing Pod. However, May refused and remained determined to go out on the field as well to join her teammates, despite Enoch warning her against it. Enoch noticed that May did not seem in her normal state, as her emotions seemed to be off.[1]

Since Enoch had been unable to convince Melinda May to remain inside Zephyr One, he followed her as she prepared to leave the plane and threatened to incapacitate her with an I.C.E.R. However, May remained unfazed by Enoch and swiftly disarmed her before carrying on. Enoch thus attempted to block her path by jumping in front of her, claiming that he had no desire to hurt her but expressing his intent to stop her.
As the fight was inevitable, May attacked Enoch, who displayed considerable combat abilities which clearly enabled him to go toe-to-toe with May, explaining that he had upgraded himself with the same package of fighting skills as the Chronicom Hunters. May remained unimpressed, stating that Enoch was still just a machine, and continued fighting Enoch, exchanging some violent blows with the Chronicom, the two opponents being able to heavily strike each other.
May was eventually able to get the upper hand on Enoch. She flipped back from the stairs of the Zephyr One's cargo ramp, landing behind Enoch and hitting his head on the stairs. Enoch attempted to retaliate, but May, who was able to begin opening the cargo ramp, trapped his leg under a seat. She then grabbed a fire extinguisher and repeately smashed Enoch's head with it, damaging his head without any restraint.
Enoch was nearly killed, but the fight was interrupted when S.H.I.E.L.D. returned from their mission, with Phil Coulson's LMD ordering May to stand down. Although she did not know about him, May was not impressed by his appearance, but still dropped her weapon and stopped fighting.[1]

Although his head had been damaged, Enoch quickly repaired his body and claimed that he was still perfectly fit for field missions. He thus joined S.H.I.E.L.D. for a rescue mission, which would eventually cause him to get stranded in 1931.
Melinda May willingly accepted to return into the Healing Pod until her healing was complete.[1] She was eventually allowed to return to field work, but discovered that she was unable to feel any emotion at all and instead felt the emotions of those she touched, which explained how ruthless she had acted towards Enoch during their fight.[3]
