Lillian Luthor is the overarching antagonist of the CW's 2015-2021 TV series Supergirl.
She is a scientist, former leader of Project Cadmus, the biological mother of Lex Luthor, the adoptive mother of Lena Luthor, and the widow of Lionel Luthor.
Lillian was portrayed by Brenda Strong in Supergirl. An alternate version of Lillian was portrayed by Nesta Chapman in Superman & Lois.
Lillian met her future husband Lionel Luthor while working as a physician. Some time after they married, they had a son, whom they named Lex. During this time, however, Lionel began to have an affair with Elizabeth Walsh. Lionel, seeking to patch things up with Lillian, paid the woman, pregnant at the time, to keep quiet about their relationship. The woman eventually gave birth to Lionel's second child, Lena.
When Lena was four years old, her mother died and Lionel decided to bring Lena to live with them. Lillian begrudgingly agreed to this and she and Lionel adopted Lena, never revealing that she was actually Lionel's biological daughter.
After Lex was arrested by his crimes against humanity, Lillian blamed Superman and all the aliens for what happened to her son and eventually joined Project Cadmus, a research lab tasked with experimenting on extraterrestrial threats. When Lillian became leader of Cadmus, she transformed it into a terrorist organization with one goal, to exterminate any alien life form on Earth. Using Cadmus' resources, she was also able to make enhancements to human bodies, as shown with Hank Henshaw/Cyborg Superman and John Corben/Metallo.

In the comics, Lillian and Leticia (Lex's mother) were reintroduced as separate characters in the Post-Flashpoint continuity (Earth-0/Prime Earth). In this version, Lillian is Lionel Luthor's sister Leticia is Lex and Lena's biological mother. They first appeared in flashbacks in Superwoman #6 (2017). According to Lex, when his parents died, he moved to Metropolis and created the greatest company on the world; LexCorp. After he left, Lillian raised Lena and encouraged her interest in science.

Viewing aliens as a poison to Earth, Lillian beings who are subtly taking control of the planet by making humans worship them, Lillian was determined to purge Earth of these otherworldly-forms. To that end, she takes over Project Cadmus, a secret lab that studies aliens, and severed its relationship with the government.
However, Lillian's arrogance makes her short-sighted (as she doesn't realize when someone close to her is manipulating or deceiving her); when her adoptive daughter, Lena apparently joined forces with her to release a deadly virus that would wipe out all alien life on earth. However, Lena had no intention of releasing the virus; she switched a critical ingredient to make the virus inert, and alerted the police about her mother's crimes. Lillian only realized she'd been double-crossed when the missile carrying the virus exploded and only sent red rain falling on National City. Before she could react, the police swarmed in and arrested Lillian for attempted mass murder and other Cadmus-related crimes.

Powers via Lexosuit: Lex Luthor's Warsuit is capable of enhancing the wearer's strength, speed, agility, and endurance.
Superhuman strength: When inside the armor, Lillian's physical strength was amplified, allowing her to lift great amounts of weight.
Superhuman speed: Lillian's armor has allowed her to reach immense speeds.
Flight: One of the armor's most important features was its propulsion system and subsequent ability of flight.
Energy Projection: The armor was able to release blasts of energy from its gauntlets.

Despite her behavior toward Lena, it is revealed in the penultimate episode of season four that Lillian does, in fact, love her, though it took having a baby Truth-Seeker alien attached to her arm to get her to admit that.
