In the prelude, he is first seen with Mary Jane Watson running over some symbiotes; they get out of the car, and Luke tells MJ that they have to go. Spider-Man then tells MJ that he was so worried; Luke Cage then tells him he best plays it cool; Luke Cage then tells MJ that they have to get on board the S.H.I.E.L.D. hovercraft. When MJ sees Symbiote Black Cat, she tells Spider-Man to get away; Luke then grabs MJ to get her to the hovercraft. They then get on board with it flying away; when the game flashbacks 4 days ago, when the Park Aves started shooting at Spider-Man, Luke Cage appears and knocks out one of the thugs. The thugs then shoot at Luke Cage, however, Luke Cage is bullet proof, with even fresh explosion of the car not hurting him at all. Luke Cage then tells the thugs that was rude; Luke Cage then helps Spider-Man take down the thugs. Luke Cage then asks Spider-Man what brings him uptown, Spider-Man then tells him; Luke Cage then tells Spider-Man, good job shutting down the drive-by, then tells him that he has been chasing these guys around all day, then tells Spider-Man that if he sees anymore, shut them down, Spider-Man agrees, then Luke Cage tells him to meet him at the park later.

After Spider-Man stops five criminals from the Rolling 7's and Park Aves, Spider-Man meets Luke at the park; Luke then tells Spider-Man, good job and that thanks to him, the gangs are thinking twice before having a shootout in the middle of the street, but they have to keep up the pressure. Spider-Man then tells Luke to tell him about these gangs; Luke then tells Spider-Man that they are engaging in a number of activities dangerous to the community and that his unique skills could be perfect for stopping them. Spider-Man then asks Luke what he meant when he said he had unique skills, Luke then tells Spider-Man he has the potential to be a great fighter but needs to use all of his senses. Spider-Man then tells Luke that he has more sense than him, Luke Cage then tells him to excuse him, Spider-Man then tells Luke then tells him about his Spider-Sense, Luke Cage then tells Spider-Man to use his Spider-Sense to its full potential, Spider-Man asks what he means. Luke Cage then tells him that he can see all of his opponents before they see him and figure out who he takes out first. Luke Cage then tells Spider-Man that he needs that exact skill right now; Spider-Man then tells Luke Cage what he needs. Luke Cage then tells Spider-Man that he has got an informant in serious danger, there's a hit about to him down, and he has no way to spot the shooter until it's too late, but Spider-Man. Spider-Man then says that he could use his Spider-Sense as a sniper-sense, Luke Cage then says exactly, and Spider-Man can bet there'll be more than one, now he will tell him where the informant is and then Spider-Man can stake it out, Spider-Man agrees.

Spider-Man heads to the location and stops the Rolling Sevens from killing the informant; Spider-Man then returns to Luke, and Luke tells Spider-Man that he did a good job and he really helped him out. Luke tells Spider-Man to use Spider-Sense to find more gangsters out on the street and take them out before they cause any trouble; Spider-Man agrees. Spider-Man does that, and Spider-Man then goes back to Luke Cage. Luke Cage tells Spider-Man not to forget that Spider-Sense will always give him the advantage; Luke Cage then tells Spider-Man that he has been watching him fight, then tells him he has tons of potential but no skills; Spider-Man then tells him that he said before he had unique skills. Luke Cage then tells Spider-Man that he has been going toe-to-toe with everybody, and with his speed, he should be coming quickly and getting out. Spider-Man then asks him what he means, and Luke Cage then offers to train Spider-Man if he is willing to learn. Spider-Man agrees, and then they go to a location. Luke Cage teaches Spider-Man all about the Web-Strike, Spider-Man learns it, Luke Cage tells Spider-Man to the show gangs what he has got and come back to him when he has mastered this, and that he will teach him some more, after Spider-Man Web-Strikes 10 thugs. Spider-Man heads back to Luke Cage; Luke Cage then tells Spider-Man, nice going and that the web stuff is working for him; Spider-Man then tells Luke it is sort of his trademark; Luke then tells him it is also sort of weird. but it's working for you, Spider-Man then thanks Luke, Luke Cage then asks Spider-Man if he is ready for some more training, Spider-Man agrees.

Spider-Man then meets Luke Cage on a rooftop; Luke Cage then tells Spider-Man that he has the perfect skill to stop the extortion these gangs have been running, then tells him that these thugs will stand in front of every door on his street, intimidating the good folks that live there into paying potential money, but Spider-Man can use his speed to hit one after the other, jump at one guy to the next, Spider-Man said he will try. Luke Cage then tells him that he can start right here with these thugs, hit him with a chain of those web attacks, Spider-Man then takes down the 3 enemies. Spider-Man and Luke Cage head to another rooftop; Luke Cage finds another group, and then Spider-Man does the same thing to the five enemies. Spider-Man and Luke Cage then head to another location, 7 of the bad guys this time. Spider-Man then asks Luke Cage if these guys do this racket all day and night. Luke Cage then says pretty much, yeah; Spider-Man then takes out Seven Thugs. Luke Cage then tells Spider-Man that he might be the answer to his problems; with all of the pressure on the gangs, maybe the good folks around here could take back the street. Spider-Man then tells Luke that all they did is push them out for a day, Luke says true, and that they need a parlay with the two gang leaders, Spider-Man tinny asks Luke how they will pull that off without them killing each other. Luke then tells them that these guys revolve around reputations and who did what to whom or for whom, then tells Spider-Man to do them a favor, then tells Spider-Man that the Park Avenues is going to try to take out the leader of the Rolling Sevens, follow the cars on the way to make the hit, stop then the Sevens will owe him, Spider-Man agrees.

Spider-Man then stops the Park Aves by destroying the five cars, stops the gas station from blowing up, and saves the Rolling 7's leader; Spider-Man then goes back to Luke Cage. Luke Cage then tells Spider-Man that it is good to see him, and that he has been thinking about Spider-Man's unique fighting skills. Spider-Man then tells him of course he has, they are awesome, Luke Cage agrees. Luke Cage then tells Spider-Man that he could use his webs to swing into a large group of enemies and them out with one single kick, Spider-Man tells Luke that they should find a large clump of thugs and that he all give it a shot. They then had to go to Central Park, where Luke Cage tells Spider-Man that this corner is infamous for gang trouble. Spider-Man then asks Luke Cage what he wants him to do, Like Cage then tells Spider-Man to get a running start and swing, and take them out with one big swinging kick, and if they spot him, they are going to try to run, so be fast. Spider-Man then succeeds; Luke Cage finds more thugs. Luke Cage then asks Spider-Man if he thinks he can do it again. Spider-Man says of course; Luke Cage then says good and tells him that it looks like they have a lookout this time, so take them out fast. Spider-Man succeeds; Luke Cage then finds another group, a big one; they have more than one lookout; just move fast and aim that swing carefully, and he should be able to get them out in one shot; Spider-Man succeeds. Luke Cage then tells him that he has nice moves, Luke Cage then tells Spider-Man to take out any groups of thugs he finds in the same way, catch them all with one swing kick, then tells him to come see him when he has cleaned up these street corners.

Spider-Man then heads to Luke Cage, Luke Cage then tells Spider-Man that he will be pleased that he heard from the leader of the Rolling Sevens, telling him that he agreed to a parley. After Luke points out that Spider-Man saved him, Luke Cage then tells Spider-Man to meet with the Park Avenue leader and tells him that the other side agrees to come to the park tonight at sundown, no guns; Spider-Man agrees, then eventually finds the leader of the Park Avenues. The Park Avenue leader agreed to it. Luke Cage is then seen with the two gang leaders getting inside the building in the park, which is unknown to them. Kingpin's men are there to assassinate the gang leaders, Spider-Man takes them down though, the Rolling Sevens leader then tells Luke Cage that he is all talk, every hero he met just used him for a punching bag, Luke Cage then tells Spider-Man that they had a deal. The Park Avenue leader then says that he has to do what he has to do, with or without me. His crew is taken down by the Sevens, and the two gang leaders then get their guns out.

Luke Cage then tells them that he said no guns, (in the Red path, Luke Cage would grab the two leaders, with Spider-Man showing them that this whole gang war was a setup from the pictures, the two gang leaders then shake hands, and then redeem themselves, telling Spider-Man if they ever need the Rolling Sevens and Park Avenues, they are there), (in the Dark path, Spider-Man would let them fight each other with their minions coming to fight, Spider-Man then takes down all 8 of the thugs, Spider-Man then says the streets are going to be save gentlemen. Luke Cage gets mad at Spider-Man, telling him what he is doing. This solves nothing, Spider-Man then tells him that this actually solves everything, then tells him that this gang war was a setup; Luke Cage then asks Spider-Man if he knew this whole turf war was a setup. Spider-Man then tells him that he asked him that he asked him to clean up the streets and that is exactly what he did, maybe they will all share a cell. Later, Luke Cage is mentioned to be with MJ. Later, when Spider-Man calls MJ again, MJ says that she has been helping Luke Cage with a shelter.

later on, during the symbiote invasion, Spider-Man meets Luke Cage and MJ at the park; Spider-man tells Luke Cage that this is pointless and that too many civilians are dying. Luke Cage then tells Spider-Man that he is doing all he can and that they can't just walk out to S.H.I.E.L.D. headquarters. Luke Cage then tells Spider-Man that he excepts all of these to fight their way to the other side of Manhattan, Luke Cage then says this is suicide. Then Spider-Man tells Luke Cage that he has to get to Wolverine, and tells him to just take care of MJ for him before MJ says that she can take care of herself just fine and that she still has friends out there, then says that she and Luke are going to them and then they are going to get out of here, Luke Cage agrees, Spider-Man then tells them to be careful, then leaves.

After Spider-Man defeats Symbiote Wolverine, Spider-Man then meets with Luke Cage and MJ, (in the Red path, Luke Cage tells Spider-Man that he is glad to see him. Luke Cage then says that they will spread thin, now he has to take the lead convoy and MJ is taking a ride with him, then, asks Spider-Man if he can protect the area until they are all under way and then guard the last convoy, then tells Spider-Man, good luck, Luke Cage and MJ then leave) (in the Dark path, Luke Cage tells Spider-Man that he has has got some nerve coming down here, then asks Spider-Man what his story is, why is he here, convoys are ready, make sure they get off safely and guard the rear, Spider-Man then tells him he can handle this whole thing by himself, Luke Cage then leaves, MJ goes with him), they went somewhere unknown, it then shows us the prelude, in the Red path, Luke Cage is last seen alongside Moon Knight and Wolverine fighting the symbiotes on top the S.H.I.E.L.D. helicarrier.
