English: Three-dimensional NT image demonstrates magnitude of CD2 tip accumulation and presence of multiple NTs. A three-dimensional image of YTS-CD2-GFP cells conjugated with 721 cells for 30 minutes on anti-CD48 coated plates. Initial image shows a single z-plane with the 721 cell outlined in white and all cells labeled with text, followed by the three-dimensional image. The transition from the two, to three-dimensional sequence is noted by a “fade-out” effect. The three-dimensional image highlights that two-dimensional imaging fails to recognize the total number of NTs. It also highlights the large accumulation of CD2 at the tip of the long NT (scale bar = 9 µm). |
