The Golden Globe Award for Best Actress - Motion Picture - Drama pertama kali diberikan oleh Hollywood Foreign Press Association sebagai kategori tersendiri sejak tahun 1951. Sebelumnya, hanya ada satu kategori, yaitu "Best Actress in a Motion Picture". Kini setelah pemisahan tersebut terdapat juga kategori Best Actress - Musical or Comedy.

Nama kategori ini sudah berubah berkali-kali sejak awal, dan mulai tahun 2005 resmi disebut: "Best Performance by an Actress in a Motion Picture-Drama".
